Themaintenanceofan intactmembranestructure isofgreat importanceforbacteria to execute various biological functions. However, chemical probesformonitoringthedynamic changesofbacterialmembranes are barely reported. Herein, we,forthefirst time, report a novelpolarity-sensitiveprobeforreflectingthepackingdegreeofbacterialmembranelipids. Specifically, we synthesize amembrane-targetingfluorescentprobe(TICT-lipid) that possesses both twist intramolecular charge transfer and aggregation-induced emission properties. TICT-lipid exhibits sensitive responses totheminute difference inthepackingdegreeofmembranelipids, facilitating rapid differentiationofGram-negative and Gram-positive bacteria. Interestingly, inthepresenceofmembrane-disrupting antibiotics,thelocalizationofTICT-lipid shifts fromtheoutermembranetothecellmembraneby outputting blue-shifted and enhanced emission, makingthemechanismofactionofantibiotics clearly visible. TICT-lipid is apolarity-sensitivefluorescentprobe, holding great promise inthestudyofmembrane-relatedbacterialprocesses and antibiotic screening.
